High Mobility Group Protein B1 (HMGB1) also known as High mobility group box 1 or SBP-1 is protein encoded by HMGB1 gene. HMGB1 is secreted by immune cells such as monocytes, macrophages, and dendritic cells through leaderless secretory pathway. Activated macrophages and monocytes secrete HMGB1. High Mobility Group Box protein 1 (HMGB1) is a non-alkaline (non-histonic) DNA-binding protein, it is normally localized in the nucleus as part of the chromatin. High Mobility Group Protein B1 is member of chromatin proteins which also includes, HMG1, HMG 2, HMG 14, and HMG 17.
HMGB1 with its high positive charge binds to DNA leads to oxidation, hyperacetylation and posttranslational modifications. HMGB1 is responsible for Stabilization of the nucleosomal structure and regulation of gene transcription in chromatin. It is released by necrotic cells hence act as chaperone to shuttle nuclei acids into the endosomal compartment of APCs. Nucleic acid sensors like TLR9 trigger due to High Mobility Group Protein B1 (HMGB1) . HMGB1 also has the ability to bind LPS, at the level of lipid A, possibly because of its high positive electric charge. It can chaperone to the CD14-TLR4 MD2 complex, increasing the response of macrophages to the LPS.
