Competent cells incorporate foreign DNA through altered cell walls to pass DNA more easily. Calcium chloride and Heat shock treatment are used to make cell “competent”. Electroporation is a process that includes brief exposure of cells to an electric field in order to pass DNA through it. Cells undergoing very rapid growth can be made competent easily than cells in other development phase. Bacterial culture has varies growth rate, in initial hours called lag phase with small number of dividing cells growth rate is low. In rapid growth phase also called Log phase under proper temperature cells can become competent easily.

Natural Competence
Cells are able to receive DNA from their environment by :
• Conjugation
• transformation
• transduction
In transformation the DNA is directly pass into the cell. In order to receive transforming DNA recipient cells should be in a specialized physiological state called log phase or competent state.
Artificial Competence
Artificial competence includes setting specific artificial conditions in which cells are made permeable to DNA in laboratory. It is relatively easy competence procedure and can be used in the genetic engineering of bacteria. Artificial competence is low efficiency process as cells are forces to adopt foreign DNA.
